Temporal Dissection of T-bet Functions

T-bet is a transcription factor of the T-box family that regulates the expression of numerous immune system-associated genes. In this report the authors produced an ectopically controllable form of T-bet by fusing its C-termial domain with a mutated ligand -binding domain of human estrogen receptor alpha. Using this temporally controllable expression system, they demonstrated that IFN-gamma, CD122 and CxR3 are all direct gene targets of T-bet.
In this study, the ImageStream system was used to demonstrate the subcellular localization of T-bet_ER before and after stimulation with 4-hydroxytamoxifen.

Mitotic Index Multiplexed with Apoptotic Cell Detection
Estimation of the mitotic index, the fraction of the cell population that is in mitosis, is an important and commonly performed assay that usually requires visual examination under a microscope. The low frequency of mitotic cells combined with manual inspection makes the assessment lengthy, tedious and often inaccurate.
The ImageStream system provides a valuable means of measuring the mitotic index of a cell population quickly and accurately. Using high resolution multispectral cell image analysis, the ImageStream can identify and quantitate cell sub-populations based on complex morphological
features. In this application, mitotic cells are identified and distinguished from apoptotic cells.
Because the ImageStream generates cell image data at high speed, a typical analysis can be carried out easily on tens of thousands of cells. These large numbers help
ensure the statistical accuracy of the results. |

MultiLaser Options for the ImageStream system
Now, you can extend the range of fluorescent dyes available to the ImageStream system by adding up to two additional solid state lasers to the ImageStream's standard 488nm laser. The new laser options include a red 658nm laser (90mW) and your choice of either a high power violet 405nm laser (350mW) or a UV 375nm laser (18mW).
